Description

Principle of the assay: mouse SAA ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for SAA has been precoated onto 96-well plates. Standards(E.coli, G20-Y122) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SAA is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse SAA amount of sample captured in plate.
Background: Serum amyloid A protein (SAA), also known as SAA1, is a protein that in humans is encoded by the SAA1 gene. This gene encodes a member of the serum amyloid A family of apolipoproteins. It is mapped to 11p15.1. SAA is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. This protein also plays an important role in HDL metabolism and cholesterol homeostasis. High levels of this protein are associated with chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, Alzheimer's disease and Crohn's disease. This protein may also be a potential biomarker for certain tumors
